In CUT&Tag, following overnight incubation with primary antibody, tubes were placed on a magnetic rack. Images show tubes before (Panel A) or after (Panel B) removal of supernatant. The sample processed without Wash Buffer Enhancers shows cloudy supernatant, poorly defined pellet boundaries, and increased sample adherence to tube walls, indicating bead aggregation and inefficient magnetic bead separation. The sample with CUTANA™ Wash Buffer Enhancer Set shows sharply defined pellet formation, minimal supernatant cloudiness, and reduced adherence to plastic surfaces, reflecting the improved bead dispersion and handling enabled by the Wash Buffer Enhancers.
EpiCypher recommends including the CUTANA™ Wash Buffer Enhancers (1 and 2, both included in this set) in CUT&RUN and CUT&Tag for optimal assay performance.
CUT&RUN
- If using a CUTANA™ CUT&RUN Kit, follow the Kit User Manual Version 5. Include Wash Buffer Enhancer 1 and Wash Buffer Enhancer 2 each at 1X final concentration in Wash Buffer.
- If using pAG-MNase (EpiCypher 15-1016) and related CUT&RUN products, follow the CUTANA™ CUT&RUN DIY Protocol. Include Wash Buffer Enhancer 1 and Wash Buffer Enhancer 2 each at 1X final concentration in Wash Buffer.
CUT&Tag
- NOTE: CUTANA™ CUT&Tag Kit Version 4 already includes Wash Buffer Enhancers 1 and 2.
- If using a CUTANA™ CUT&Tag Kit (Version 3 or prior), follow the Kit User Manual Version 3. Include Wash Buffer Enhancer 1 and Wash Buffer Enhancer 2 each at 1X final concentration in Wash Buffer 1.
- If using pAG-Tn5 (EpiCypher 15-1017) and related CUT&Tag products, follow the CUTANA™ CUT&Tag DIY Protocol. Include Wash Buffer Enhancer 1 and Wash Buffer Enhancer 2 each at 1X final concentration in Digitonin150 Buffer and Digitonin300 Buffer.
