Description

This product differs from previous lots due to the addition of an HA tag and TEV spacer between the N-terminal His tag and the bromodomain. Recombinant human BRD4 Bromodomain 1, His-Tagged, expressed in E. coli (HUNK1, accession number O60885, amino acids 41 to 180). BRD4 is a member of the BET sub-family of bromodomain-containing proteins, which also includes BRD2, BRD3 and BRDT. BET proteins have two bromodomains and an ET domain C-terminal to the second bromodomain. BRD4 serves as a co-factor for Myc and is crucial to Myc’s role in acute myeloid leukemia. Inhibition of BRD4 binding suppresses Myc activity and leads to differentiation of AML cells and elimination of AML stem cells.

Recombinant 6xHis-HA-tagged protein at 0.98 mg/mL in 20 mM Tris-HCl pH 8.0, 200 mM NaCl, 1 mM DTT, 20% glycerol.
Storage and Stability
Stable for six months at -80°C from date of receipt. For best results, aliquot and avoid multiple freeze/thaws.

Application Notes

Recombinant BRD4 Bromodomain 1, His-HA-tagged is useful for protein binding and screening experiments examining acetylated protein substrates.
