EpiCypher specializes in synthesis of multiply modified peptides containing post-translational modifications (PTMs), unnatural amino acids, and fluorescent tags. We use automated Fmoc Solid Phase Peptide Synthesis (SPPS) to make the synthetic peptides, routinely making 25-30 amino acid peptides. Generally, 50-mer peptide synthesis is considered the upper limit for standard SPPS, but the real limits are highly dependent on the peptide sequence and required purity level.
Peptides are generally synthesized from 10 to 100 µmol scale (~5 to 200 mg of crude peptide), but large scale synthesis can be performed if needed.
Peptide Quality Control:
Peptide homogeneity is evaluated by mass analysis (MALDI-TOF mass spectrometry) and analytical HPLC.
Below are some of the peptide modifications we can introduce into your peptide sequence:
- Acetylation (N-terminal or Lys(Ac)
- Biotinylation methylation (on Lys or Arg residues)
- Phosphorylation (on Ser, Thr, or Tyr residues)
- Fluorescent tags (e.g 5-Fam , 5(6)-Tamra)
- D-forms of amino acids
- Unnatural amino acids (e.g. 6-Cl-Trp)
- Amino acids containing stable isotopes (e.g. 2H, 13C, 15N)
- PEG linkers/spacers
EpiCypher can also make Multiple Antigenic Peptides (MAPs) for polyclonal antibody generation. If we do not have a non-standard (modified or unnatural) amino acid in the lab, we can order it (users will be charged for the price of the order). We would be happy to discuss your peptide needs and other modifications that you may require. We also have the capability to synthesize mid-sized libraries of crude peptides (~100 peptides in 2-3 weeks).
